Macrophage podosomes assemble at the leading lamella by growth and fragmentation.

Using high resolution 3D and 4D light microscopy, Evans et al investigate the assembly and turnover of the dynamic adhesion structures in macrophages called podosomes. 3D kymography was used to quantify the effects of cytoskeleton-perturbing drugs on podosome dynamics and longevity.
